If you have an aftermarket soundcard, and onboard, make sure the onboard is disabled in BIOS. Also, i have a Soundblaster X-Fi Platinum, and on the Input side of things, you have to manually select which input you want to use (i'm running Vista 32, TMK XP doesn't require it to be set manually, and i've no idea why vista does, it should just allow multiple inputs, and control over each..)

When i originally plugged my mic in, i had to select it as the default input, and Reboot.

Ever since then it has been fine. Hope this helps

Another possible solution is that the front panel connectors for the audio are not connected properly to your motherboard. If you built it yourself you may have forgotten to connect these cables, or they may have been dislodged due to moving the PC case.
Imma try deactiving the onboard later...i jsut installed the other soundcard because onbord strangely stopped working....
