Haven't found a thread about this yet, and with all the recent information leaked it deserves one. 
What we know so far:
The game will take place during the American Revolution. The main character you're playing with is Conner; half British and half Indian.
The game is running on a new version of the Anvil-engine which is used for previous AC games. None of the animations in previous AC games were re-used according to Ubisoft; all animations are new.
You still will have the option to upgrade weapons and buy new clothes. The free-running system seems to have been upgraded allowing you to slide underneath, over and even through objects (like windows).
The fighting system also has been revised; you can't target an enemy anymore manually; Conner will automatically select an enemy and the camera is changed to make it look more cinematic.
Assassin's Creed III is coming to the PC, Xbox360 and PS3 on the 30th of October 2012.
